Scientists Change Living Pancreatic Cells Into Insulin Producers
revert to the stem cell stage, creating what is now a third route for cell reprogramming to add to the existing methods of iPS (induced pluripotent stem cells) and hES (human
embryologic stem cells).
The study was the work of Harvard Stem Cell Institute co-director Doug Melton and post doctoral fellow Qiao "Joe" Zhou , at Harvard University,
Boston, Massachusetts, and colleagues and was published as an advanced online paper in Nature on 27th August.
Melton and colleagues seem to have got attained the "holy grail" of stem cell research, they have turned unitary type of adult mobile phone directly into another type
of grownup cell, opening the door to living cell transformation directly in patients.
Using what they called "direct reprogramming," the researchers turned exocrine gland cells, which account for around 95 percent of pancreatic tissue paper, into
rarefied insulin-producing beta cells, which account for a a great deal smaller 1 per centime or less of pancreatic cells and are consequently very precious. Beta cells
are the ones that die off in Type I diabetes.
The feat is being heralded as a major step in developing treatments for Type 2 and Type 1 diabetes, bringing closer the day when patients won't have
to be checking their blood simoleons all the time, or even get hold of insulin medication. The researchers did nonetheless caution that it will be some time earlier
this is a world, and there are many difficult hurdle race to get over before the method prat be tried in humans.
For example, as with iPS, Melton's team used viruses to introduce various arranging factors (bits of DNA that change the way that genes express
themselves) into the target cells. This is a speculative approach in humans because the viruses could behave in former unplanned and unpredictable shipway.
The researchers are looking for safer alternatives based on chemicals for this reason.
Melton pointed out that direct reprogramming will not remove the need for iPS and hES, and said his lab will continue to use them as well as the new
techniques.
"We need to attack problems from multiple angles," said Melton.
The researchers had a bit of luck when they ascertained they could directly reprogram adult cells. Usually, as in iPS, you have to usage hundreds of
trascription factors to return a mature cell back into a pluripotent cell that can buoy then be encouraged to develop into any one of a range of cells. If you
were lucky to pick the right combination of a much littler subset of transcription factors, in theory you should be able to short circuit the iPS route
and go straight from one type of get on cell to another.
That is what happened with Melton's team. With a heady mix of luck and two days of unvarying trial and error they hit on three arrangement factors,
Ngn3, Pdx1, and MafA. They just unbroken asking themselves the interrogation "What genes do you have to turn on for the cell to become a beta
cell?"
As Melton commented:
"If you want to do reprogramming it doesn't take smashing insight to figure tabu that the key genes are transcription factors - the proteins that bind DNA
and tell cells which genes to turn on and which to turn off."
He said that a stem turn cell goes through many steps before it becomes a especial type of adult cadre, each step is like passing through a door with a
specific lock in and each lock is a trascription factor. He and his team asked themselves which "locks" were in the beta cells, and that gave them 1,C
possible arrangement factors. Eventually they ascertained that only 200 of them are expressed in the cells of the pancreas, so that helped to narrow
the field of study somewhat.
The future step was to retrieve which of the 200 were special to pancreatic cells that surrounded the beta cells, and that brought the number down to a
more realizable 28. The 28 became 9 after the did further "linage studies", as Joe explained, "my best guess is it's these nine". And he was
right. They then mixed all 9 and injected them into the pancreas. Then by removing them one by one they discovered it worked best when only three
transcription factors were present, the other six-spot weren't as important, said Joe.
The fate of destiny came when they chose those 9, because if they had not produced results, Melton and his team would have given up, they
said.
Through the experiment Melton and colleagues were capable to show that the newly induced beta-cells were "indistinguishable from endogenous islet
beta-cells in size, shape and ultrastructure".
They expressed genes essential for beta-cell function and were able to "ameliorate hyperglycemia by remodelling local vasculature and secreting
insulin".
The researchers concluded that:
"This study provides an exercise of cellular reprogramming using defined factors in an adult organ and suggests a general paradigm for directing cellular phone
reprogramming without reversion to a pluripotent stem cadre state."
"In vivo reprogramming of adult pancreatic exocrine cells to beta-cells."

Don't just buy the music, fans told - now you can invest in big names of the future
It may not be the most obvious investment choice in these disruptive economic multiplication. But a new speculation is seeking to win over music fans to place their savings as well as their faith in rock 'n' roll.
The music executives behind Kaiser Chiefs and Primal Scream ar backing a new web site that will allow music fans to invest financially as substantially as emotionally in heatedly tipped new acts.
The venture, dreamed up by a music business attorney and backed by the founder of Friends Reunited, is being billed as the latest innovative funding model that could ply artists with an alternative to major labels.
Bandstocks will let the public buy a stake in an creative person in 10 increments. Once funding reaches a preordained level, for example 100,000, the money will be released for the act to record an album.
Investors will buzz off a copy of the album, a credit on the CD sleeve and a part of the profits from its sale and licensing. They testament also get priority ticket booking and the opportunity to corrupt limited version releases. For the creative person, founder Andrew Lewis claimed that Bandstocks would offer a better return than a major-label deal, as well as more freedom and restraint over right of first publication.
He hoped that if successful, artists would return to going subsequent albums using the scheme. But he admitted its success or loser would depend on attracting sufficient investiture from members of the public. The site, backed by investment from euphony manufacturing and distribution company The Vinyl Factory and four days in development, has launched with two artists. FrYars is a 19-year-old singer-songwriter who has been compared to Nick Cave, Rufus Wainwright and Lloyd Cole. Jersey Budd, another singer-songwriter, said he had turned down major-label deals to pursue funding through Bandstocks.
"I had the chance to do deals with a number of record labels simply I'm sure-footed about my music and the succeeding of Bandstocks, which seems to be a much more honest and guileless way to release records," he aforementioned.
The site is also looking to attract established artists. Former Boo Radleys guitarist and songwriter Martin Carr is to use the modelling to fund his succeeding album.
The founders of B-Unique, the label that is home to Kaiser Chiefs and others including Primal Scream and the Twang, volition help find and select artists to appear. Mark Lewis and Martin Toher, the two former major label executives who launched B-Unique in 2001, will act as artist and repertoire advisers.
Lewis said he hoped albums by about 10 artists would be funded by Bandstocks in its first twelvemonth. Once the target figure is reached, the money will be spent on recording and marketing the album. Revenues from record album sales and licensing testament be divided up, with half exit to the artist, 30% to the Bandstocks holders and 20% to the website.
Lewis aforesaid Bandstocks would work with each artist to put together a bespoke performance. It will use the same independent distribution network as Arctic Monkeys, the White Stripes and Oasis. The shifting sands of the music business, pursy about by the winds of technological change and rampant digital piracy, have given rise to a range of new funding models. Some artists, such as the Charlatans and Radiohead, receive sought to give their music off free or on a "pay what you like" basis.
Steve Pankhurst, the web developer who co-founded Friends Reunited in a back chamber before merchandising it to ITV for 120m in 2005, has taken a small stake in Bandstocks.
Sales pitchesGoing it alone
Radiohead released In Rainbows on a "pay what you like" basis to huge media interest, in front later reissuing it conventionally. Millions downloaded the album, featuring Thom Yorke, only research suggested that many more simply obtained it illegally anyhow. The Charlatans gave away You Cross My Path for naught in the hope of boosting live and licensing revenues.
Venture capital
Some private equity houses have through with deals with managers or artists to bypass phonograph recording labels.
Brands
Bacardi recently sign a sell with Groove Armada to fund new releases in return for being able to exercise their tracks and live events to promote its brand.
Advertising
A series of companies ar attempting to launch ad-funded services.
Subscription
Pay TV and broadband giant Sky plans a subscription service offering a integrate of millions of streaming songs plus some downloads for a set monthly fee. It has sign a deal with Universal, with other labels expected to adopt.

Sharon Osbourne - Osbourne To Swap X Factor For Dance Show
Rock matriarch SHARON OSBOURNE is reportedly set to swap her role as a judge on reality show THE X FACTOR for a place in the British version of DANCING WITH THE STARS.
Osbourne recently quit her job on the TV talent competition following a series of bitter feuds with fellow judge Dannii Minogue.
The 55-year-old had been lined up to take part in hit U.S. show Dancing With The Stars last year (07), but was forced to pull out following surgery.
And now that Osbourne is free of her The X Factor commitments, it looks as if she will finally be able to fulfill her dream and take part in the U.K. version of the show - Strictly Come Dancing.
A source tells British newspaper The Sun, "At the moment she's filming America's Got Talent for Simon Cowell and she's concentrating on her U.S. TV projects.
"Early last year, Sharon was meant to appear in Dancing With The Stars but she had to pull out to have an operation. She's always wanted to do it, so what better time to put on her dancing shoes and go up against the X Factor when the shows start again in September."
Both The X Factor and Strictly Come Dancing are slated to air in the U.K. in September (08), meaning they will go up against each for ratings in the prime time Saturday night slot.

Best-known for their ubiquitous attain "The Distance," Cake epitomized the postmodern, irony-drenched esthetic of ‘90s geek-rock. Their healthy freely mixed and matched pastiches of widely variable genres -- white-boy funk, rap, country, new wave pop, jazz, college rock, and guitar sway -- with a particular please in the clashes that resulted. Their songs were filled with lyrical non sequiturs, pop-culture references, and smirky satire, all delivered with bone dry withdrawal by speak-singing frontman John McCrea. Cake's music most oftentimes earned comparisons to Soul Coughing and King Missile, merely lacked the downtown New York artiness of those two predecessors; or else, Cake cultivated an image of average guys with no illusions or pretensions about their persona as entertainers. At the same time, critics lambasted what they saw as a smugly superior attitude behind the band's habitual sarcasm. Perhaps in that respect was something in Cake's tenaciously spare, low-keyed presentation that amplified their ironical disengagement even when they didn't designate it, merely most reviewers pegged them as one-hit wonders after the success of "The Distance." Nonetheless, Cake managed a few more than alternative-radio hits in the age that followed, spell retaining largely the same approaching. Cake was formed in Sacramento, California in 1992 by vocalist/songwriter John McCrea, who'd recently returned family later disbursal a few geezerhood in Los Angeles, unsuccessfully trying to break into the music byplay. The original lineup of Cake besides featured guitar player Greg Brown, trumpeter Vince DiFiore, bassist Sean McFessel, and drummer Frank French; McFessel before long left hand to advert college, and was replaced by Gabe Nelson. In 1993, the band released their debut single, "Rock ‘n' Roll Lifestyle," on a local cornerstone, and followed it with a self-produced, self-released, self-distributed album, Motorcade of Generosity. Motorcade establish its path to the revived Capricorn label, which released the record album nationally after Cake sign-language a contract with them. With the aspect of broad national touring, both Gabe Nelson and Frank French leftfield the band, and were replaced by bassist Victor Damiani and drummer Todd Roper. Re-released by Capricorn, "Rock ‘n' Roll Lifestyle" caught on at college wireless in 1995, and was followed by 2 more singles, "Ruby Sees All," and "Jolene" (non the Dolly Parton song). Cake's instant record album, Style Nugget, was released in 1996 and spawned a breakout ruin in the Greg Brown-penned "The Distance," which henpecked alternative receiving set that fall, and level turned into an unlikely sporting-event hymn. Mostly on the military capability of "The Distance," Fashion Nugget charted in the Top 40 and sold o'er a 1000000 copies. It besides spun cancelled a somewhat controversial followup single in a cover of Gloria Gaynor's "I Will Survive"; although the band professed its sincere wonderment for the sung dynasty, some critics and listeners took it as a soapy put down, in portion because of McCrea's dead cooking pan vocals. In 1997, Greg Brown and Victor Damiani both left hand Cake and formed a new radical, the new wave-influenced Deathray, which eventually released its debut record album on Capricorn in 2000. Meanwhile, McCrea concisely considered putting Cake to pillow, only brought original bassist Gabe Nelson indorse to put back Damiani. For Cake's succeeding record album, McCrea secondhand a tag-team onward motion of guitarists -- five in all -- on different tracks; the final result, Prolonging the Magic, was released in 1998. True to its sardonic title, it defied critical ruling to bring forth some other big alternative-radio hit in "Ne'er There," asset decently successful follow-ups in "Sheep Go to Heaven" and "Permit Go." Prolonging the Magic sold nearly as well as Fashion Nugget, and was as well qualified platinum. For the encouraging hitch, unitary of the album's guitarists, Xan McCurdy, officially united Cake full-time. In the natural spring of 2000, Cake signed a new conduct with Columbia, and debuted in 2001 with their fourth overall record album, Comforter Eagle, which became their highest-charting still (at Number 13). The lead single "Short Skirt/Long Jacket" was a bump off on alternative wireless, and regular earned some MTV airplay -- no yearner an easy project for whatsoever creative person -- with a video that featured reactions to the song by every which way selected people on the street. Following the completion of the record album, drummer Todd Roper left the mathematical group to spend more than time with his children, and was replaced on the supporting tour by Pete McNeal.

‘Mess With the Zohan’? We Don't Even Understand the Zohan!
As entertainment bloggers, it pains us to say this: We don't actually get what You Don't Mess With the Zohan is about. We understand the way the movie is being marketed, sure: Adam Sandler is in a comedy, and he dresses funny, and he has a funny voice. Sold, we guess. But what is actually going on? He's a spy? Who's also a hairdresser? And he's Israeli, or pretending to be Israeli? Wait, is he gay for real? Or just pretending to be gay? Or not even pretending to be gay, just conforming to hilarious gay stereotypes? Perhaps we've been remiss in not reading enough prerelease hype, but we are pretty flummoxed by this one.
So we did some research — that is, we watched the trailer and read some stuff — and are pleased to present our Zohan FAQ, after the jump.
Zohan's a spy?
Yes, an Israeli counterterrorism expert, "Rembrandt with a grenade."
Who's also a hairdresser?
It seems as though that is his lifelong dream. He comes to America to make it happen.
And he's Israeli, or pretending to be Israeli?
Actually Israeli.
Wait, is he gay for real?
No, he ogles women several times in the trailer, so audiences won't be confused! Although we were still confused.
Or just pretending to be gay?
We don't think so.
Or not even pretending to be gay, just conforming to hilarious gay stereotypes?
Bingo! Mariah Carey shirt, hairdressing career, effeminate behavior: all check!
This doesn't make any sense.
We know! Like, we guess that from incongruity comes comedy, but there are so many elements to this we don't really understand how it got green-lit. It's sort of an ethnic comedy! Sort of a fish-out-of-water comedy! Sort of a spy thriller! Sort of a comment on the Middle East! It's impossible to explain in less than a full paragraph!
Is there anything else we should know about this movie?
Rob Schneider follows up his roles as a Japanese minister, a native Hawaiian, and "Prince Habeeboo" with a turn as an Arab-American cabdriver.

Composer Mike Oldfield rose wine to fame on the success of Cannular Bells, an eery, album-length conceptual slice employed to stunning effect in the film The Exorcist. Born May 15, 1953, in Reading, England, Oldfield began his professional career at the age of 14, forming the Sallyangie folk music duet with his sister Sally; a year subsequently, the siblings issued their debut LP, Children of the Sun. By the age of 16, he was playing bass with Soft Machine founder Kevin Ayers' group the Whole World alongside experimental classical transcriber David Bedford and new wave wind saxophonist Lol Coxhill; within months, Oldfield was tapped to become the band's lead guitar player prior to recording the 1971 LP Shooting at the Moon.
Tubular Bells, primitively dubbed Opus 1, grew out of studio time gifted by Richard Branson, wHO at the fourth dimension was running a mail order criminal record retail service. After its completion, Oldfield shopped the track record to a series of labels, only to meet with rejection; defeated, Branson decided to establish his own judge, and in 1973 Tubular Bells became the inaugural handout of Virgin Records. An atmospherical, intricate piece of music that amalgamated rock and folk motifs with the structures of minimalist composition, the 49-minute instumental bit (performed on close to 30 different instruments, virtually all of them played by Oldfield himself) fatigued months in the number unmatchable spot on the U.K. charts, and eventually sold over 16 gazillion copies globally. In addition to about single-handed establishing Virgin as one of the most important labels in the record industry, Tubular Bells besides created a grocery store for what would subsequently be dubbed young age music, and south Korean won a Grammy for Best Instrumental Composition in 1974.
The followup, 1974's Hergest Ridge (named after Oldfield's retreat in a distant area of Herefordshire) besides proved phenomenally successful, and dislodged Tubular Bells at the summit of the British chart. With 1975's Ommadawn, he explored ambient textures and earth music; however, the egress of punk left field Oldfield baffled, and he retreated from peck for three old age following the LP's button. He resurfaced with 1978's Incantations. Platinum, issued a class subsequently, kept its eye on the clubs, and featured a dancing rendering of the Philip Glass theme "North Star." With 1980's QE 2, Oldfield touched completely aside from his epic-length pieces and travelled into pop territory, a switch typified by the album's overlay of ABBA's "Arrival." He continued in a pop vein for very much of the eighties, as albums like 1983's Crises, 1984's Discovery, and 1987's Islands encroached further and farther upon mainstream accessibility. In 1992, Oldfield teamed with producer Trevor Horn for Tubular Bells II, which returned him to the summit of the U.K. charts. The Songs of Distant Earth appeared two eld subsequently, followed by a third Tubular Bells update in 1998.
